    Summary

This study presents a new fluorimetric method for measuring adrenaline and noradrenaline in urine using ion exchange chromatography. The enhanced trihydroxyindole method offers stable fluorescence and accurate results, even with alpha-methyldopa use.

    Area of Science:

    • Biochemistry
    • Analytical Chemistry

    Context:

    • Accurate quantification of catecholamines is crucial for diagnosing various medical conditions.
    • Existing methods for catecholamine analysis often face challenges with sensitivity, specificity, or stability.

    Purpose:

    • To develop and validate a novel, sensitive, and stable fluorimetric method for the simultaneous determination of adrenaline and noradrenaline in urine.
    • To establish reference ranges for urinary catecholamines in healthy individuals.
    • To assess the method's reliability in the presence of alpha-methyldopa, a common antihypertensive medication.

    Summary:

    • A new trihydroxyindole fluorimetric method was employed for catecholamine separation using ion exchange chromatography.
    • Enhanced fluorescence stability and differentiation of adrenaline and noradrenaline were achieved through specific chemical treatments (boric acid, copper-ions, mercaptoethanol, reacidification) and pH-controlled oxidation.
    • The method demonstrated high precision, accuracy, sensitivity, and specificity, with normal values established for healthy adults and validated in patients with primary hypertension treated with alpha-methyldopa.

    Impact:

    • Provides a reliable and enhanced analytical method for routine clinical assessment of catecholamine levels.
    • Contributes to improved diagnostic capabilities for conditions related to catecholamine imbalances.
    • Offers a robust assay applicable in research and clinical settings, unaffected by common therapeutic interventions like alpha-methyldopa.
