We developed new water-soluble BODIPY dyes for cell membrane imaging. These dyes exhibit far-red absorption and near-infrared fluorescence, enabling effective plasma membrane labeling with high photostability.
Area of Science:
- Organic Chemistry
- Biophysical Chemistry
- Cell Biology
Background:
- BODIPY dyes are widely used fluorescent probes.
- Developing water-soluble variants with specific cellular localization is challenging.
- Far-red/near-infrared probes are desirable for reduced autofluorescence and deeper tissue penetration.
Purpose of the Study:
- To synthesize novel water-soluble BODIPY dyes.
- To achieve specific labeling of the cell plasma membrane.
- To investigate their photophysical properties and stability.
Main Methods:
- Synthesis of dicationic and dianionic BODIPY derivatives.
- Spectroscopic characterization (absorption, emission).
- Cellular imaging studies to assess localization and photostability.
Main Results:
- Two water-soluble BODIPY dyes with far-red absorption and NIR fluorescence were synthesized.
- Dicationic and dianionic groups ensured water solubility and plasma membrane localization.
- The dicationic dye showed excellent plasma membrane localization and resistance to photobleaching (>8 min).
- Dyes were nonemissive in polar solvents but highly fluorescent in nonpolar solvents and upon membrane insertion.
Conclusions:
- Novel water-soluble BODIPY dyes enable effective and photostable plasma membrane labeling.
- The dicationic derivative is a promising probe for live-cell imaging of the plasma membrane.
- These dyes offer advantages for biological imaging due to their spectral properties and localization.

Physical Properties Affecting Solubility
As for any solution, the solubility of a gas in a liquid is affected by the attractive intermolecular forces between solute and solvent species. Unlike solid and liquid solutes, however, there is no solute-solute intermolecular attraction to overcome when a gaseous solute dissolves in a liquid solvent since the atoms or molecules comprising a gas are far separated and experience negligible interactions. Consequently, solute-solvent interactions are the sole...
Solubility
A solution is a homogeneous mixture composed of a solvent, the major component, and a solute, the minor component. The physical state of a solution—solid, liquid, or gas—is typically the same as that of the solvent. Solute concentrations are often described with qualitative terms such as dilute (of relatively low concentration) and concentrated (of relatively high concentration).
In a solution, the solute particles (molecules,...
