Background:

  • Large arteries are composed of elastin, collagen, and muscle, observable via light and transmission electron microscopy.
  • Elastin forms the internal elastic lamina (IEL) in all arteries and fenestrated sheets within the media of large arteries like the aorta.

Purpose of the Study:

  • To describe the structural organization of elastin within large arteries.
  • To investigate the relationship between arterial elastin and the development of atherosclerosis.

Main Methods:

  • Light microscopy and transmission electron microscopy were employed to visualize arterial components.
  • Chemical treatment (0.1 N NaOH at 70-75°C for 5 hours) was used to isolate adventitial elastin and vasa vasorum.

Main Results:

  • Fenestrations in the medial elastic sheets are larger than those in the IEL.
  • Adventitial elastin is fibrous and surrounds vasa vasorum.
  • Fenestration size increases near arterial branches and in post-stenotic dilatations.

Conclusions:

  • Atherosclerosis is associated with dynamic changes in elastin, including new formation in early stages and elastolysis in advanced disease.
  • The structural characteristics of elastin in large arteries may play a role in the pathogenesis of atherosclerosis.
