New 2,4-dinitrobenzenesulfonyl derivatives rapidly release phenols upon thiol trigger. Derivative 1 induces DNA cross-linking via glutathione-mediated quinone methide formation.
Area of Science:
- Organic Chemistry
- Medicinal Chemistry
Background:
- 2,4-dinitrobenzenesulfonyl derivatives are useful chemical reagents.
- Thiol-responsive compounds are valuable in chemical biology and drug delivery.
Purpose of the Study:
- To synthesize novel 2,4-dinitrobenzenesulfonyl derivatives.
- To investigate their reactivity with thiols.
- To explore their potential for inducing DNA cross-linking.
Main Methods:
- Simple synthetic procedures for preparing derivatives 1-3.
- Kinetic studies using proton nuclear magnetic resonance (¹H NMR) and high-performance liquid chromatography (HPLC) to monitor thiol-triggered release.
- Investigation of DNA cross-linking induced by derivative 1 in the presence of glutathione.
Main Results:
- Three new 2,4-dinitrobenzenesulfonyl derivatives (1-3) were synthesized.
- Derivatives 1-3 rapidly released corresponding phenol derivatives (4-6) within 5 minutes upon exposure to thiols (glutathione, L-cysteine).
- Derivative 1 demonstrated the ability to induce DNA cross-linking in the presence of glutathione, suggesting a mechanism involving quinone methide formation.
Conclusions:
- The synthesized 2,4-dinitrobenzenesulfonyl derivatives exhibit rapid thiol responsiveness.
- The observed DNA cross-linking capability of derivative 1 highlights its potential as a therapeutic or research tool.
