Summary

Synovial fluid cells from inflammatory arthritis patients exhibit high phospholipase A2 (PLA2) activity. This enzyme can also passively adsorb to peripheral blood neutrophil membranes from synovial fluid, indicating a potential source of inflammation.

Background:

  • High phospholipase A2 (PLA2) activity is observed in synovial fluids (SF) of inflammatory arthritides.

Purpose of the Study:

  • To investigate the presence and origin of PLA2 within SF cells.
  • To determine if PLA2 can adsorb to peripheral blood neutrophils.

Main Methods:

  • PLA2 content was measured in sonicates of SF cells and peripheral blood neutrophils (PMN).
  • Spontaneous release of PLA2 from unstimulated cells was assessed.
  • PMN were incubated with SF or purified PLA2 to evaluate passive adsorption.

  • SF cells showed significantly higher PLA2 activity compared to normal peripheral blood PMN.
