Notch1 signaling protects heart cells from injury by maintaining mitochondrial quality control. Overexpression of Notch1 improved cell viability and mitochondrial function during ischemia reperfusion injury.
Area of Science:
- Cardiovascular Biology
- Mitochondrial Biology
- Cellular Signaling
Background:
- Mitochondrial quality control is crucial for myocardial protection.
- Notch signaling is vital for heart development and repair.
- The role of Notch in myocardial mitochondrial quality control is not well understood.
Purpose of the Study:
- To investigate the role of Notch1 in myocardial mitochondrial quality control following ischemia reperfusion injury (IRI).
- To elucidate the protective mechanisms of Notch1 in the heart.
Main Methods:
- Isolated rat myocardial cells and induced an IRI model.
- Modulated Notch1 expression using recombinant adenoviruses (Ad-N1ICD for overexpression, Ad-shN1ICD for knockdown).
- Assessed cell viability, ATP production, mitochondrial fusion/fission dynamics, mitophagy, and key protein phosphorylation (Pink1, Mfn2, Parkin).
Main Results:
- Ischemia reperfusion injury reduced myocardial cell viability.
- Notch1 overexpression enhanced cell viability, ATP production, and mitochondrial fusion while decreasing fission and mitophagy.
- Notch1 knockdown produced opposite effects.
- Notch1-mediated protection involved inhibiting Pink1 expression and phosphorylation of Mfn2 and Parkin.
Conclusions:
- Notch1 exerts a protective effect on the myocardium.
- This protection is linked to the maintenance of mitochondrial quality control.
- Notch1 inhibits the Pink1/Mfn2/Parkin signaling pathway to preserve mitochondrial function during injury.
