Microarray expression profile and analysis of circular RNA regulatory network in malignant pleural effusion

Circular RNAs (circRNAs) are newly identified in lung adenocarcinoma-associated malignant pleural effusion (LA-MPE). These circRNAs may play a role in LA-MPE development by regulating cancer-related pathways.

Purpose of the Study:

- Quantitative real-time PCR (qRT-PCR) was employed to validate the expression of top upregulated circRNAs.
- CircRNA-miRNA-mRNA networks were constructed, and pathway analysis was performed.
Main Results:
- 1350 differentially expressed circRNAs and 1727 differentially expressed mRNAs were identified in LA-MPE versus TPE.
- Four of the top five upregulated circRNAs were validated by qRT-PCR, with hsa_circ_0085386 showing an increasing trend.
- Pathway analysis revealed that aberrantly expressed circRNAs potentially regulate key cancer-related pathways, including PI3K-Akt signaling and Axon guidance.
Conclusions:
- This study provides the first specific circRNA profiles for LA-MPE.
- Aberrantly expressed circRNAs may contribute to LA-MPE development by influencing cancer-related signaling pathways through miRNA sequestration.
