Tumor necrosis factor receptor-associated factor 3 (TRAF3) is vital for type I interferon induction. Its function in host defense against viral infections is cell-type and stimulus-specific, with TRAF2 playing a similar role.
Area of Science:
- Immunology
- Molecular Biology
- Virology
Background:
- Tumor necrosis factor receptor-associated factor 3 (TRAF3) mediates type I interferon (IFN) induction via pattern recognition receptors.
- The in vivo role of TRAF3 in antiviral defense remains unclear due to limited mouse models.
Purpose of the Study:
- To investigate the in vivo function of TRAF3 in host defense against viral infections.
- To elucidate the cell type- and stimulus-specific roles of TRAF3 and TRAF2 in type I IFN induction.
Main Methods:
- Conditional and inducible knockout mouse models for TRAF3 deletion in specific cell types (myeloid cells, dendritic cells) and whole body.
- Vesicular stomatitis virus (VSV) infection model.
- Analysis of type I IFN induction in various cell types (MEFs, macrophages, dendritic cells) upon stimulation with TLR ligands or viruses.
Main Results:
- Conditional TRAF3 deletion in myeloid or dendritic cells did not significantly impair host defense against VSV.
- Whole-body inducible TRAF3 deletion increased susceptibility to VSV infection.
- TRAF3 was essential for type I IFN induction in MEFs but not macrophages; its role in dendritic cells was ligand-dependent.
- TRAF2 was also found to be essential for type I IFN induction across multiple cell types.
Conclusions:
- TRAF3 and TRAF2 are crucial for type I IFN induction.
- The functions of TRAF3 and TRAF2 in IFN induction are specific to cell type and stimulus.
