A new variant of Tomato leaf curl New Delhi virus (ToLCNDV) was identified in cucumber plants in Indonesia. This finding highlights the need for farmers to manage whitefly vectors to prevent begomovirus spread in cucurbits.
Area of Science:
- Plant virology
- Molecular biology
- Agricultural science
Background:
- Cucumber (Cucumis sativus L.) is a vital vegetable crop in Indonesia.
- Yellowy green mosaic symptoms were observed on cucumber leaves in Central Java in 2008.
Purpose of the Study:
- To identify the viral pathogen causing mosaic symptoms in Indonesian cucumber plants.
- To characterize the genomic sequences of the identified virus.
Main Methods:
- DNA extraction from symptomatic cucumber leaves.
- Polymerase Chain Reaction (PCR) amplification using specific primers for viral genomic DNA.
- DNA sequencing of amplified viral DNA-A and DNA-B components.
Main Results:
- Full-length DNA-A and DNA-B sequences of a bipartite virus were obtained.
- The virus isolate showed high nucleotide sequence identity (95.5% for DNA-A, 91.0% for DNA-B) to Tomato leaf curl New Delhi virus-[cucumber:Thailand].
- The virus was identified as a variant of ToLCNDV and named ToLCNDV-Indonesia[Indonesia:Java:Cucumber:2008] (ToLCNDV-ID[ID:Jav:Cuc:08]).
Conclusions:
- This is the first report of Tomato leaf curl New Delhi virus naturally occurring in Indonesia.
- The study identifies a new begomovirus affecting cucumber in Indonesia.
- Farmers are advised to increase vigilance in controlling whiteflies, the vectors of begomoviruses, particularly on cucurbit crops.
