SEMA4D shows promise as a prognostic biomarker for various cancers, improving survival predictions. However, it was not associated with breast cancer prognosis, unlike its receptor Plexin-B1 in Caucasian patients.
Area of Science:
- Oncology
- Biomarker Discovery
- Molecular Biology
Background:
- SEMA4D and its receptor Plexin-B1 have demonstrated potential in predicting carcinoma patient prognosis.
- Previous studies suggest a link between SEMA4D/Plexin-B1 expression and cancer outcomes.
Purpose of the Study:
- To conduct a meta-analysis evaluating the prognostic role of SEMA4D and Plexin-B1 in various malignancies.
- To determine the association between SEMA4D/Plexin-B1 expression and patient survival across different cancer types.
Main Methods:
- Systematic literature search across PubMed, Embase, Web of Science, and CNKI databases.
- Extraction of Hazard Ratios (HRs) and 95% Confidence Intervals (CIs) from 14 eligible studies.
- Meta-analysis including 2638 patients to assess prognostic relevance.
Main Results:
- High SEMA4D expression significantly correlated with overall survival (OS) and disease-free survival (DFS/PFS/RFS) in tumors (HR=2.05, HR=1.59).
- No significant association was found between SEMA4D expression and breast cancer prognosis (HR=0.76).
- Plexin-B1 expression positively correlated with OS and DFS in Caucasian breast cancer patients (HR=0.56, HR=0.68).
Conclusions:
- SEMA4D may serve as a prospective prognostic biomarker for numerous malignancies, excluding breast cancer.
- Plexin-B1 shows significant prognostic value for Caucasian breast cancer patients.
- Further research may elucidate the specific roles of SEMA4D and Plexin-B1 in distinct cancer types.
