The Wald-Wolfowitz runs test, commonly referred to as the runs test, is a nonparametric test used to assess the randomness of ordered data. The test evaluates the number of runs, which are consecutive sequences of similar elements within the data.   • Lipopolysaccharide (LPS) is a potent inducer of systemic inflammation.

Purpose of the Study:

  • To investigate the impact of LPS-induced inflammation on glucose/insulin tolerance, exercise performance, and neurotransmitter levels in mice.
  • To determine if moderate physical exercise can mitigate the adverse effects of LPS-induced inflammation.

Main Methods:

  • Adult male C57BL/6 mice were subjected to LPS injections to induce systemic inflammation.
  • Glucose and insulin tolerance tests were performed.
  • Exercise performance was assessed using an incremental loading test.
  • Urinary neopterin levels and striatal neurotransmitter concentrations (dopamine, serotonin) were measured.
  • A moderate treadmill exercise protocol was implemented for three weeks.

Main Results:

  • Repeated LPS administration impaired glucose tolerance, increased urinary neopterin, and reduced striatal dopamine levels.
  • LPS also increased the serotonin/dopamine ratio and decreased exercise performance.
  • Moderate exercise training prevented the exacerbation of immune activation (indicated by neopterin levels).
  • Exercise preserved mitochondrial activity in skeletal muscle despite continuous LPS infusion.

Conclusions:

  • Peripheral inflammation induced by LPS causes metabolic disturbances and affects striatal dopamine metabolism.
  • Moderate physical exercise serves as a protective intervention against LPS-induced inflammation.
  • Exercise mitigates immune system activation and maintains mitochondrial function in inflammatory states.
