Background:

  • Coffee (Coffea arabica L.) varieties 'Caturra' and 'Catuaí' exhibited severe symptoms in Costa Rica starting in 1995.
  • Symptoms included reduced leaf size, malformed and curling leaves, shortened internodes, chlorotic mosaic, necrosis, flower/bean abortion, and reduced yield, termed 'crespera'.

Purpose of the Study:

  • To identify the causal agent of the 'crespera' disease affecting coffee plants in Costa Rica.
  • To determine the geographic distribution and potential vectors of the identified pathogen.

Main Methods:

  • Grafting experiments were conducted using infected and healthy budwoods on coffee plants.
  • Enzyme-linked immunosorbent assay (ELISA) was used to test field-collected symptomatic plants and greenhouse-inoculated plants for X. fastidiosa.
  • Leafhopper insects found in affected areas were collected and tested for the presence of X. fastidiosa using ELISA.

Main Results:

  • Grafting experiments confirmed the transmission of symptoms to healthy plants.
  • ELISA tests revealed 100% of symptomatic field samples and 3 of 10 grafted plants were positive for X. fastidiosa.
  • X. fastidiosa was detected in leafhopper species Graphocephala permagna (34.5%) and Erythrogonia sonora (23.8%), though vector status requires further investigation.

Conclusions:

  • Xylella fastidiosa is confirmed as the causal agent of the 'crespera' disease in coffee in Costa Rica.
  • The study extends the known geographic distribution of X. fastidiosa to Costa Rica.
  • The findings highlight the need for further research into potential insect vectors and disease management strategies.
