Fibroblast growth factor-2 (FGF-2) enhanced bone regeneration and angiogenesis in irradiated rat calvarial defects. Hyperbaric oxygen (HBO) therapy improved angiogenesis but had minimal impact on bone healing.
Area of Science:
- Regenerative Medicine
- Biomaterials Science
- Surgical Oncology
Background:
- Radiotherapy can impair bone healing, particularly in craniofacial reconstructions.
- Fibroblast growth factor-2 (FGF-2) and hyperbaric oxygen (HBO) therapy are potential therapeutic modalities for enhancing tissue repair.
- Biphasic calcium phosphate (BCP) is a commonly used bone graft material.
Purpose of the Study:
- To histologically evaluate bone regeneration in irradiated rat calvarial defects.
- To assess the efficacy of FGF-2 and BCP blocks, with or without HBO therapy, in promoting bone healing.
Main Methods:
- Irradiated calvarial defects were created in rats and treated with FGF-2 or BCP blocks.
- Animals were divided into HBO and non-HBO (NHBO) groups.
- Defects were analyzed radiographically and histologically after 4 weeks of healing.
Main Results:
- FGF-2 significantly enhanced new bone formation compared to controls.
- BCP blocks showed no improvement in bone regeneration.
- HBO therapy showed a trend towards increased angiogenesis but minimal effect on bone formation.
- FGF-2 efficacy was not significantly improved by adjunctive HBO therapy.
Conclusions:
- FGF-2 promotes bone regeneration and angiogenesis in irradiated calvarial defects.
- HBO therapy benefits angiogenesis but has limited impact on bone regeneration in this model.
- The combination of FGF-2 and HBO did not yield superior results compared to FGF-2 alone.
