Advanced-age mice show protection against diet-induced obesity, unlike younger mice. However, they experienced increased liver fat accumulation and inflammation when fed a Western diet (WD).

  • Male C57BL/6JRj mice of two age groups (10 weeks and 22 months) were fed either a WD or a standard rodent diet (SFD) for 15 weeks.
  • Body weight, adipose tissue (AT) mass, plasma adiponectin and leptin levels were measured.
  • Hepatic lipid accumulation and macrophage marker F4/80 expression were assessed.
  • mRNA levels of uncoupling protein-1 in brown adipose tissue (BAT) and intestinal integrity markers were analyzed.

Main Results:

  • Advanced-age mice on a WD did not exhibit increased body weight or AT mass, contrasting with young mice.
  • Plasma adiponectin and leptin levels remained unaffected by WD feeding in advanced-age mice.
  • WD feeding induced greater hepatic lipid accumulation and increased hepatic F4/80 expression in advanced-age mice.
  • No significant changes in BAT activity or intestinal integrity markers were observed, suggesting these are not responsible for the protective effect.

Conclusions:

  • Advanced-age mice appear protected against diet-induced obesity compared to younger mice.
  • Despite protection from obesity, advanced-age mice show increased liver steatosis and inflammation under WD conditions.
  • The mechanisms behind obesity protection in aged mice warrant further investigation.
