Genetic variations in PHLDB1 are linked to nonfunctional pituitary adenoma (NFPA) risk in the Korean population. A specific single-nucleotide polymorphism, rs67307131, showed a significant association with increased NFPA susceptibility.
Area of Science:
- Genetics
- Oncology
- Endocrinology
Background:
- Previous studies linked PHLDB1 single-nucleotide polymorphisms (SNPs) to glioma risk.
- The association between PHLDB1 SNPs and pituitary adenoma risk remained uninvestigated.
Purpose of the Study:
- To evaluate the association between PHLDB1 SNPs and the risk of developing pituitary adenomas.
- Specifically investigate the link between PHLDB1 genetic variants and nonfunctional pituitary adenoma (NFPA) risk.
Main Methods:
- A case-control study was conducted in the Korean population.
- Genotyped 27 PHLDB1 tagging and exon SNPs in 148 NFPA patients and 375 controls.
- Logistic regression analysis was employed to assess the association between PHLDB1 SNPs and NFPA risk.
Main Results:
- A significant association was detected between a PHLDB1 SNP (rs67307131) and NFPA risk.
- The 'C/T' genotype of rs67307131 demonstrated a higher odds ratio (OR=2.39) and a stronger association (P=0.00002).
- Functional analysis predicted rs67307131 to be a CpG site and copy number variant, potentially influencing disease susceptibility.
Conclusions:
- Genetic variations in PHLDB1 may be associated with the risk of NFPA.
- This study is the first to report an association between PHLDB1 variants and NFPA.
- Further research is warranted to confirm the impact of rs67307131 on NFPA susceptibility.
