Background:

Purpose of the Study:

  • To evaluate the reliability and consistency of canine serum IgE testing using monoclonal anti-canine enzyme-linked immunosorbent assay (ELISA).
  • To assess the impact of sample handling and storage time on test results and interpretation.

Main Methods:

  • Serum samples from 35 atopic dogs were divided into three aliquots.
  • Two aliquots (Samples 1 and 2) were tested immediately; the third (Sample 3) was stored at -80°C for approximately 30 days before testing.
  • Specific IgE reactivity to various allergens was measured using a monoclonal anti-canine ELISA.

Main Results:

  • High variability in IgE reactivity percentages was observed between samples tested on the same day (14.30% to 127.34%) and samples tested a month apart (21.78% to 129.65%).
  • Interpretation differences occurred in 15.18% of allergens between same-day samples, affecting 91.4% of dogs.
  • Interpretation differences increased to 22.32% when comparing samples stored for a month, with all dogs showing at least one differently interpreted allergen.

Conclusions:

  • Canine allergen-specific IgE serology testing demonstrates significant variability, potentially altering immunotherapy treatment plans.
  • There is a critical need to improve the reliability and standardization of monoclonal anti-canine ELISA for IgE testing in veterinary diagnostics.
