A novel bacterium, Roseovarius faecimaris, was discovered in a Korean tidal flat. This obligate aerobic, Gram-negative bacterium expands the known diversity within the Roseovarius genus.
Area of Science:
- Microbiology
- Bacterial Taxonomy
- Marine Biology
Background:
- The Roseobacter clade, within the Rhodobacteraceae family, comprises diverse marine bacteria with significant ecological roles.
- Understanding bacterial diversity in marine environments like tidal flats is crucial for ecological studies.
Purpose of the Study:
- To isolate and characterize a novel bacterium from a tidal flat environment.
- To determine the taxonomic placement of the novel isolate using polyphasic characterization.
Main Methods:
- Isolation and cultivation of strain MME-070T from a tidal flat.
- Physiological and biochemical characterization, including growth conditions and substrate utilization.
- Phylogenetic analysis based on 16S rRNA gene sequencing.
- Analysis of cellular fatty acids, polar lipids, and respiratory quinones.
- Determination of genomic DNA G+C content.
Main Results:
- Strain MME-070T is a Gram-negative, obligate aerobic, non-motile rod.
- Optimal growth occurred at 25–30°C, pH 8.0, with 2–3% NaCl.
- 16S rRNA gene sequence analysis showed 96.9% similarity to Roseovarius aestuarii SMK-122T.
- Phylogenetic analysis placed the isolate within the genus Roseovarius.
- Chemotaxonomic data (Q-10, major fatty acids) supported its placement in the genus.
Conclusions:
- Strain MME-070T represents a novel species within the genus Roseovarius.
- The proposed name for the novel species is Roseovarius faecimaris sp. nov.
- This discovery contributes to the understanding of Roseobacter diversity in coastal marine ecosystems.
