    Summary

    Psoriatic skin shows reduced 15-hydroxyeicosatetraenoic acid (15-HETE) production in the dermis. This defect in 15-HETE generation may contribute to the development of psoriasis by allowing increased inflammatory lipoxygenase products.

    Background:

    • Psoriatic lesions exhibit increased levels of 5-lipoxygenase and 12-lipoxygenase products.
    • 15-hydroxyeicosatetraenoic acid (15-HETE) normally inhibits these inflammatory lipoxygenase pathways.

    Purpose of the Study:

    • To investigate the in vitro capacity of dermis from uninvolved psoriatic skin to produce 15-HETE.
    • To compare 15-HETE production in psoriatic dermis versus normal dermis.

    Main Methods:

    • Homogenized dermis from uninvolved psoriatic and normal skin was used.
    • Lipid extraction followed by reversed-phase high-performance liquid chromatography (HPLC).
    • Identification of lipid peaks using authentic standards, UV spectrometry, and radioimmunoassay.

    Conclusions:

    • Defective 15-HETE generation in the dermis of psoriatic skin may be a significant factor in psoriasis development.
    • Reduced 15-HETE could lead to increased inflammatory and proliferative lipoxygenase products, exacerbating psoriatic pathology.
