    Summary

    Paracetamol elixir absorption and metabolism are similar whether given orally or via a jejunostomy tube. This finding supports the effective use of jejunostomy administration for patients needing this route.

    Background:

    • Paracetamol (acetaminophen) is a common analgesic.
    • Administration via jejunostomy tube is used for patients unable to take oral medication.
    • Understanding paracetamol absorption via jejunostomy is crucial for effective pain management.

    Purpose of the Study:

    • To compare the pharmacokinetic disposition of paracetamol oral elixir administered via feeding jejunostomy versus oral administration.
    • To evaluate the absorption and biotransformation of paracetamol in patients receiving it through a jejunostomy tube.

    Main Methods:

    • Two male patients received 650 mg paracetamol elixir via feeding jejunostomy.
    • Plasma concentration-time data were analyzed for area under the curve, half-life, and time to maximum concentration.
    • Urinary metabolites (paracetamol glucuronide and sulphate) were quantified and compared to four oral control subjects.

    • Area under the plasma concentration-time curve, elimination half-life, and time to maximum concentration were similar between jejunostomy and oral administration groups.
