Molecular and Ionic Solids

20.0K
Crystalline solids are divided into four types: molecular, ionic, metallic, and covalent network based on the type of constituent units and their interparticle interactions.
Molecular Solids
Molecular crystalline solids, such as ice, sucrose (table sugar), and iodine, are solids that are composed of neutral molecules as their constituent units. These molecules are held together by weak intermolecular forces such as London dispersion forces, dipole-dipole interactions, or hydrogen bonds, which...

Area of Science:

  • Ophthalmology
  • Genomics
  • Molecular Biology

Background:

  • Understanding the pathophysiology of age-related macular degeneration (AMD) at the local tissue level is limited.
  • Tissue-specific changes in gene expression, microenvironment, and epigenetics are crucial for AMD.
  • Developing new treatments for AMD requires analyzing human ocular tissues, which is challenging.

Purpose of the Study:

  • To present a standardized protocol for the rapid processing of postmortem donor eyes.
  • To enable molecular, biochemical, and genomic studies of ocular tissues for AMD research.
  • To facilitate the development of translational treatment and prevention strategies for AMD.

Main Methods:

  • Partnered with an eye bank to obtain donor eyes, blood, and vitreous within 6 hours postmortem.
  • Performed phenotypic analysis using spectral-domain optical coherence tomography (SD-OCT) and fundus photography.
  • Isolated macular and extramacular tissues, separating neural retina and retinal pigment epithelium/choroid for preservation and analysis.

Main Results:

  • Multimodal imaging provided high-resolution retinal pathology, enhancing phenotypic rigor.
  • Analysis yielded excellent quality RNA, confirming appropriate tissue segregation.
  • Ocular disease phenotype was rigorously analyzed by ophthalmologists using standardized grading criteria.

Conclusions:

  • The Utah protocol offers a standardized methodology for AMD disease mechanism analysis.
  • This protocol enables simultaneous rigorous phenotypic, molecular, biochemical, and genomic analysis of local and systemic tissues.
  • The approach is vital for developing disease biomarkers and therapeutic interventions for AMD.
