    Summary

    Interferon-gamma enhances monocyte killing of tumor cells by acting on both monocytes and cancer cells. Interferon-alpha did not show similar effects, but could be potentiated by interferon-gamma.

    Background:

    • Human peripheral blood monocytes play a role in tumor cell destruction.
    • Understanding the mechanisms of monocyte-mediated cytotoxicity is crucial for cancer therapy.

    Purpose of the Study:

    • To investigate the mechanism of human peripheral blood monocyte-mediated cytotoxicity.
    • To determine the role of interferons (IFNs) in modulating this process.

    Main Methods:

    • Utilized HT-29 colon adenocarcinoma, A673 rhabdomyosarcoma, and A375 melanoma cell lines as targets.
    • Pretreated target cells with recombinant human interferon-gamma (IFN-gamma) and interferon-alpha (IFN-alpha).
    • Assessed changes in susceptibility to monocyte killing at various effector/target cell ratios.
