    Summary

    Growth hormone-releasing hormone (GHRH) can cause acromegaly when secreted by tumors. GHRH therapy effectively stimulates growth hormone release, showing promise for treating growth hormone deficiency in children.

    Background:

    • Acromegaly is typically caused by pituitary adenomas, but can also result from ectopic Growth Hormone-Releasing Hormone (GHRH) secretion.
    • GHRH was isolated from pancreatic tumors causing clinical acromegaly.

    Purpose of the Study:

    • To investigate the effects of GHRH administration on growth hormone (GH) secretion in various populations.
    • To evaluate the therapeutic potential of GHRH for GH deficiency.

    Main Methods:

    • Administration of GHRH to normal adults, adults with childhood GH deficiency, and GH-deficient children.
    • Continuous infusion and intermittent subcutaneous administration of GHRH.
    • Intranasal administration of GHRH.

    Main Results:

    • GHRH administration stimulates GH secretion in normal adults and some GH-deficient individuals.
    • Continuous GHRH infusion augments natural GH pulses in normal men.
    • Subcutaneous and intranasal GHRH are effective, with higher doses required for intranasal delivery.
    • Intermittent subcutaneous GHRH therapy promotes linear growth in GH-deficient children.

    Conclusions:

    • Ectopic GHRH secretion is a rare cause of acromegaly.
