  • Mast cells (MCs) play a central role in allergic responses.

Purpose of the Study:

  • To investigate the microRNA profile of human mast cells during IgE-mediated sensitization and activation.
  • To correlate miRNA expression with prostaglandin D2 synthesis and histamine release.
  • To compare miRNA profiles between allergic asthmatic patients and nonatopic controls.

Main Methods:

  • Expression profiling of 762 miRNAs in mast cells derived from CD133+ stem cells.
  • Cells were isolated from allergic asthmatic patients and nonatopic controls.
  • Mast cells underwent IgE-mediated sensitization and activation.

Main Results:

  • IgE-mediated sensitization and activation significantly increased miRNA-210 expression.
  • The miRNA-132/212 cluster also increased post-activation.
  • Histamine release was higher in allergic MCs, with several miRNAs correlating to mediator release.

Conclusions:

  • The study elucidates the role of specific miRNAs in mast cell activation and mediator release.
  • miRNA-210 may be crucial in mast cell sensitization contributing to allergic symptoms.
