Purpose of the Study:

  • To develop and optimize a novel method for simultaneous enrichment and quantification of six key biogenic amines in fish samples.
  • To establish an efficient, cost-effective, and environmentally friendly analytical technique for biogenic amine analysis.

Main Methods:

  • A Stop-and-Go-Extraction Tip (StageTip)-based method was employed for simultaneous enrichment of six biogenic amines.
  • Ultrahigh-performance liquid chromatography coupled with triple quadrupole mass spectrometry (UPLC-MS) was utilized for identification and quantification without derivatization.
  • The method was optimized for short extraction time, small sample size, and minimal organic solvent consumption.

Main Results:

  • High recovery efficiencies for biogenic amines (84% to 110%) were achieved in spiked fish samples.
  • Excellent linearity (R² > 0.995) was observed for biogenic amine quantification in the range of 0.4 to 3.0 mg/L.

Conclusions:

  • The developed StageTip-based method combined with UPLC-MS provides a powerful tool for analyzing biogenic amines in complex food matrices like fish.
  • This approach is low-cost, environmentally friendly, and offers superior analytical performance compared to existing methods.
  • The method accurately reflects the subtle distribution of biogenic amines within food samples, enhancing food safety assessment.
