Background:

Glomerulotubular balance is crucial for kidney function.
This balance is maintained during development despite changing regulatory factors.
The mechanisms underlying this developmental adaptation are not fully understood.

Purpose of the Study:

To quantify changes in factors controlling glomerulotubular balance in developing guinea pigs.
To describe the profile of these adaptations in the proximal tubule epithelium.
To elucidate the mechanisms maintaining glomerulotubular balance during postnatal development.

Main Methods:

Measured hydrostatic and oncotic pressures to assess physical forces.
Estimated proximal tubule permeability using macromolecule permeance and histologic measurements.

Assessed tubular fluid osmolality (TF/Posm) at the end of the proximal tubule.

Net reabsorption pressure increased significantly from 15.0 to 30.9 mmHg between 1 and 50 days of age.
Proximal tubule permeability to mannitol decreased with age, indicating a reduction in paracellular channel size.
Tubular fluid osmolality (TF/Posm) decreased, suggesting increased solute reabsorption and tighter epithelium.

Conclusions:

Early postnatal glomerulotubular balance is facilitated by high proximal tubule permeability compensating for low reabsorption pressure.
Maturation involves a tighter epithelium, necessitating increased intercellular channels and active sodium transport for sustained balance.
These findings provide insights into kidney development and adaptation.
