Purpose of the Study:

  • To compare four extraction methods for TRPs: hot water extraction (HWE), ultrasonic-assisted extraction (UAE), enzyme-assisted extraction (EAE), and ultrasonic-enzyme-assisted extraction (UEAE).
  • To optimize the EAE method for TRP extraction using response surface methodology (RSM).

Main Methods:

  • Extraction of TRPs using HWE, UAE, EAE, and UEAE.
  • Optimization of EAE conditions including liquid-solid ratio, enzymolysis time, enzyme dosage, and pH via RSM.

  • Optimized EAE conditions (30 mL/g, 87 min, 1.6% enzyme, pH 6) achieved a maximum TRP yield of 13.15%.

Conclusions:

  • EAE is the most effective method for extracting TRPs from Trifolium repens L., offering superior yield and bioactivity.
  • Optimized EAE parameters provide a robust protocol for maximizing TRP recovery.
