A novel Streptococcus pneumoniae mutant (Δpep27) vaccine shows potential for treating asthma. Intranasal immunization with Δpep27 suppressed allergic airway disease features in mice, suggesting its therapeutic use.
Area of Science:
- Immunology
- Allergology
- Microbiology
Background:
- Asthma is an allergic airway disease (AAD) driven by immune dysregulation.
- Regulatory T cells (Tregs) are crucial for immune homeostasis.
- Streptococcus pneumoniae components and infection may induce Tregs, offering therapeutic potential for asthma.
Purpose of the Study:
- To investigate the potential of a live attenuated Streptococcus pneumoniae pep27 deletion mutant (Δpep27) vaccine for asthma protection.
- To evaluate the efficacy of intranasal Δpep27 immunization in a mouse model of allergic airway disease.
Main Methods:
- Mice were immunized intranasally with Δpep27 before or after ovalbumin sensitization and challenge.
- Hallmark features of allergic airway disease were assessed, including immune cell infiltration, cytokine production, and airway remodeling.
Main Results:
- Δpep27 immunization significantly suppressed key asthma indicators.
- This included reduced antigen-specific type 2 helper T cell responses and antibody production.
- Pulmonary eosinophil accumulation and goblet cell hyperplasia were also suppressed.
Conclusions:
- Intranasal Δpep27 immunization effectively ameliorated allergic airway disease in a mouse model.
- The Δpep27 vaccine demonstrates feasibility as a preventive or therapeutic agent for asthma.
- Further research into Δpep27's immunomodulatory mechanisms is warranted.

Asthma: Pathogenesis and Management
Asthma is classified as allergic and non-allergic. Allergens such as dust mites, pollen, and pet dander trigger allergic asthma, while factors like cold air, intense emotions, or exercise can induce non-allergic asthma.
