  • Map-based cloning was employed to isolate the UMPK gene from a rice mutant.
  • Biochemical assays were performed to measure nucleotide content (UDP, UTP).
  • Chlorophyll content, photosynthetic capacity, and gene expression (plastid and nuclear-encoded RNA polymerase-dependent genes) were analyzed.

Main Results:

  • A recessive mutant with pale-green leaves was identified, linked to a mutation in the UMPK gene.

  • Mutant plants exhibited reduced transcription of plastid-encoded genes and elevated transcription of nuclear-encoded genes, along with increased sensitivity to cold, heat, and drought stress.

  • UMPK is involved in regulating plant responses to various environmental stresses, including cold, heat, and drought.
