Human thymus leukemia-like antigens (CD1a-c) are glycoproteins found on thymocytes and dendritic cells. CD1c, detected by M241, is expressed on B cells in SCID patients, suggesting a link to murine Qa antigens.

    Area of Science:

    • Immunology
    • Cell Biology
    • Biochemistry

    Background:

    • Human thymus leukemia-like antigens (CD1a-c) are glycoproteins expressed on thymocytes, T cell leukemias, and dendritic cells.
    • CD1 molecules are hypothesized to be human counterparts of murine thymus leukemia antigens.
    • Severe combined immunodeficiency disease (SCID) affects immune cell development.

    Purpose of the Study:

    • To investigate the expression of CD1c antigen (M241) on circulating mononuclear cells in children with SCID.
    • To determine if CD1c expression is limited to B cells in SCID or also present on normal B cells.
    • To explore the relationship between CD1c and murine Qa antigens.

    Main Methods:

    • Two-color immunofluorescence analysis to detect M241 expression on mononuclear cells.

    Related Experiment Videos

  • Serological and biochemical analyses on purified B cells from various tissues.
  • Parallel analyses using monoclonal antibodies NA1/34 and 4A76 for CD1a and CD1b.
  • Main Results:

    • CD1c (M241) was expressed on 43-95% of circulating mononuclear cells in SCID children.
    • M241 expression was restricted to B cells (B4/CD19, B1/CD20, surface immunoglobulin positive).
    • CD1c expression was confirmed on purified normal B cells from spleen, tonsil, and peripheral blood; CD1a and CD1b were negative on B cells.

    Conclusions:

    • CD1c is expressed on a subset of normal peripheral B cells, not just in SCID.
    • This finding suggests CD1c may be a human analog of murine Qa antigens.
    • CD1a and CD1b are not expressed on B cells.
