1Department of Veterinary Public Health, Faculty of Agriculture, Miyazaki University, Kibanadai-Nishi, Gakuen, Miyazaki-shi 889-21, Japan.
A novel continuous separation method enables the detection and identification of nine sulfonamides using bioassay and high-performance liquid chromatography. This technique offers a reliable way to quantify these important antimicrobial agents.
Area of Science:
- Analytical Chemistry
- Microbiology
- Pharmacology
Background:
- Sulfonamides are crucial antimicrobial agents.
- Accurate determination of sulfonamide levels is essential for therapeutic efficacy and safety.
- Existing methods may lack the sensitivity or specificity required for complex mixtures.
Purpose of the Study:
- To develop and validate a continuous separation method for the simultaneous determination of nine sulfonamides.
- To establish a bioassay-guided approach for sulfonamide detection.
- To utilize high-performance liquid chromatography (HPLC) for precise identification and quantification.
Main Methods:
- A bioassay using Bacillus subtilis ATCC 6633 on minimum medium was employed for initial sulfonamide detection.
- Extracts from inhibition zones were analyzed using high-performance liquid chromatography (HPLC).
- Two distinct HPLC conditions utilizing a μ-Bondapak C18 column and an Asahipak GS-320 column were optimized for separation and identification.
Main Results:
- The method successfully separated eight to nine sulfonamide peaks depending on the HPLC column used.
- Retention times ranged from 4.68 to 50.78 min (μ-Bondapak C18) and 12.62 to 54.43 min (Asahipak GS-320).
- Standard curves exhibited excellent linearity (correlation coefficients >0.99) with recoveries between 25.2% and 64.1%.
Conclusions:
- A robust continuous separation method coupled with HPLC has been established for sulfonamide analysis.
- The developed method is suitable for the identification and quantification of multiple sulfonamides in a single run.
- This approach provides a valuable tool for quality control and research involving sulfonamide compounds.
