MiR-655-3p inhibited proliferation and migration of ovarian cancer cells by targeting RAB1A
1Department of Gynecology and Obstetrics, The People's Hospital of China Three Gorges University, Yichang, China. 609715926@qq.com.
Objective:
To investigate the potential effect of microRNA-655-3p (miR-655-3p) on the development of ovarian cancer (OC) and its relevant mechanism.
Patients And Methods:
Expression level of miR-655-3p in OC tissues was detected. The potential target gene of miR-655-3p was firstly predicted online and subsequently verified by luciferase reporter assay and Western blot. In vitro effects of miR-655-3p on SKOV3 cells were determined as well.
Results:
Low expression of miR-655-3p in OC was confirmed by quantitative Reverse Transcription-Polymerase Chain Reaction (qRT-PCR). Ras-related protein Rab-1A (RAB1A) was a direct target of miR-655-3p in OC and was negatively regulated by miR-655-3p. Further, the effects of miR-655-3p/RAB1A axis on cell proliferation, metastasis ability, and EMT activation were emphasized.
Conclusions:
Our research emphasized the suppressor function of miR-655-3p in OC. By targeting RAB1A, miR-655-3p played a tumor suppressor role in OC. We affirmed the beneficial effects of miR-655-3p in OC cells for the first time, thus providing an experimental basis for the treatment of OC.
Insights
microRNA-655-3p (miR-655-3p) acts as a tumor suppressor in ovarian cancer (OC). It targets RAB1A, inhibiting OC cell proliferation and metastasis, offering a potential therapeutic strategy.
Area of Science:
- Oncology
- Molecular Biology
- Biochemistry
Background:
- Ovarian cancer (OC) remains a leading cause of cancer-related mortality in women.
- Understanding the molecular mechanisms underlying OC development is crucial for identifying novel therapeutic targets.
- MicroRNAs (miRNAs) have emerged as critical regulators in various cancers, including OC.
Purpose of the Study:
- To investigate the role of microRNA-655-3p (miR-655-3p) in ovarian cancer pathogenesis.
- To elucidate the molecular mechanism by which miR-655-3p influences OC development.
- To explore the potential of miR-655-3p as a therapeutic agent for OC.
Main Methods:
- Quantitative Reverse Transcription-Polymerase Chain Reaction (qRT-PCR) was used to assess miR-655-3p expression levels in OC tissues.
- Bioinformatic tools predicted potential target genes of miR-655-3p, followed by validation using luciferase reporter assays and Western blotting.
- In vitro experiments were conducted to evaluate the effects of miR-655-3p on SKOV3 ovarian cancer cells, focusing on proliferation, metastasis, and epithelial-mesenchymal transition (EMT).
Main Results:
- miR-655-3p expression was found to be significantly downregulated in OC tissues compared to normal tissues.
- Ras-related protein Rab-1A (RAB1A) was identified as a direct target of miR-655-3p and was negatively regulated by it.
- The miR-655-3p/RAB1A axis was shown to suppress ovarian cancer cell proliferation, inhibit metastasis, and reduce EMT activation.
Conclusions:
- miR-655-3p functions as a tumor suppressor in ovarian cancer.
- Targeting the miR-655-3p/RAB1A pathway offers a promising therapeutic strategy for OC.
- This study provides a novel experimental basis for the development of miR-655-3p-based therapies for ovarian cancer.
