Bipolar disorder is a chronic mental health condition marked by significant mood fluctuations, including episodes of mania and depression. Elevated energy levels, heightened mood or irritability, impulsive behavior, reduced sleep needs, rapid speech, racing thoughts, inflated self-esteem, and distractibility characterize mania. Individuals with bipolar disorder often alternate between depressive and manic states, with periods of emotional stability lasting an average of six months to a year.

Summary

Structural and functional brain connectivity differences are key in bipolar disorder. Decreased structural-functional coupling strength in bipolar disorder patients may help identify individuals and inform prevention strategies.

Background:

  • Aberrant structural and functional brain connectivity are hallmarks of bipolar disorder.
  • Limited research has investigated the agreement between structural and functional connectivity (SC-FC) in bipolar disorder.

Purpose of the Study:

  • To evaluate if the structural-functional connectivity coupling index offers clinical predictive value for bipolar disorder spectrum disorders.
  • To assess whether SC-FC coupling provides additional information beyond intramodality network measures.

Main Methods:

  • Utilized diffusion tensor imaging (DTI) for structural connectivity and resting-state functional magnetic resonance imaging (rs-fMRI) for functional connectivity.
  • Analyzed network properties and SC-FC coupling index in 57 euthymic bipolar disorder patients and 42 healthy controls (ages 13-28).

Main Results:

  • Bipolar disorder patients exhibited increased structural rich-club connectivity and decreased functional modularity compared to controls.
  • Significantly decreased structural-functional connectome coupling strength was observed in patients, serving as a key discriminating feature.

Conclusions:

  • Structural-functional coupling strength is a potential valuable trait-like biomarker for bipolar disorder, surpassing intramodality network measures.
  • This measure holds promise for early identification, prevention of onset, and relapse management in bipolar disorder.
