Immunoglobulin D (IgD) and its receptor (IgDR) are overexpressed in diffuse large B-cell lymphoma (DLBCL). This abnormal activation state of IgD and IgDR suggests a potential role in DLBCL pathogenesis.
Area of Science:
- Immunology
- Oncology
- Hematology
Background:
- Immunoglobulin D (IgD) levels are elevated in autoimmune diseases.
- The role of IgD and its receptor (IgDR) in diffuse large B-cell lymphoma (DLBCL) oncogenesis is not well-defined.
Purpose of the Study:
- To investigate the expression of IgD and IgDR in DLBCL patients.
- To explore the correlation between IgD, IgDR, and clinical parameters in DLBCL.
Main Methods:
- Analysis of membrane IgD (mIgD) and IgDR in tissue samples via immunohistochemistry (IHC).
- Flow cytometry (FCM) for mIgD and IgDR expression on peripheral blood mononuclear cells (PBMCs).
- Enzyme-linked immunosorbent assay (ELISA) for secreted IgD (sIgD) levels; statistical analysis using Fisher's exact test and Spearman correlation.
Main Results:
- mIgD and IgDR were upregulated in DLBCL patients' lymph nodes and PBMCs compared to controls.
- mIgD expression correlated with IgDR expression, serum β2-microglobulin, and the Germinal Center B (GCB) phenotype.
- IgDR expression correlated with serum lactate dehydrogenase (LDH), IPI score, and GCB phenotype; sIgD levels were elevated and correlated with β2-microglobulin and LDH.
Conclusions:
- Overexpression of IgD and IgDR indicates an abnormal activation state in DLBCL.
- IgD and IgDR may play a significant role in the pathogenesis of DLBCL.
