A gene is a stretch of DNA that serves as the blueprint for functional RNAs and proteins. The structure and stability of mRNA molecules regulates gene expression, as mRNAs are a key step in the pathway from gene to protein. In eukaryotes, the half-life of mRNA varies from a few minutes up to several days. mRNA stability is essential in growth and development. The absence of the proteins regulating its stability, such as tristetraprolin in mice, can cause systemic issues, including bone marrow overgrowth, inflammation, and autoimmunity.

Background:

Oral clefts are common birth defects with variable expressivity, often attributed to epigenetic factors.
The precise molecular mechanisms of chromatin regulators like MOZ in craniofacial development remain largely unknown.

Purpose of the Study:

To investigate the role of the MOZ (KAT6A) gene in craniofacial development and its impact on gene expression during mouse development.
To identify specific gene networks regulated by MOZ that are associated with oral clefts.

Main Methods:

Utilized genetic interaction and genomic studies in mouse models to assess gene expression changes upon MOZ loss.
Analyzed differential gene expression profiles and identified genes previously linked to cleft palates.
Investigated MOZ occupancy at specific gene loci and its effect on histone acetylation.

Main Results:

Loss of MOZ resulted in differential expression of over 500 genes, including 19 associated with cleft palates.
Four distal-less homeobox (DLX) transcription factor genes (Dlx1, Dlx2, Dlx3, Dlx5) and their targets were significantly affected.
MOZ directly occupied the Dlx5 locus and was essential for histone H3 lysine 9 acetylation, regulating DLX gene expression cell-autonomously in neural crest cells.

Conclusions:

The chromatin modifier MOZ plays a critical role in regulating craniofacial development by controlling a specific gene expression program.
MOZ's regulation of DLX transcription factors and their targets provides a molecular link between chromatin modification and oral cleft development.
