Background:
Paclitaxel and docetaxel affect microtubule polymerization, yet surprising differences in tumor sensitivity to the taxanes have been observed. Docetaxel was superior to paclitaxel in inhibiting in vivo growth of human lung and prostate but not breast cancer models.
Materials And Methods:
We compared drug cytotoxicity, effects on β-tubulin isoforms, markers of apoptosis and proteomic profiles in human prostate (LNCaP), lung (SK-MES, MV-522) and breast (MCF-7, MDA-231) cancer cell lines in vitro.
Results:
Cytotoxicity followed the order SK-MES
Conclusion:
The superior activity of docetaxel in tumors with low Bcl-2 warrants further studies on biomarkers for drug sensitivity and investigation of docetaxel in combination with drugs that reduce Bcl-2 gene expression.
Insights
Docetaxel shows greater effectiveness than paclitaxel in treating lung and prostate cancers. This study explored taxane drug sensitivity, revealing Bcl-2
Area of Science:
- Oncology
- Pharmacology
- Molecular Biology
Background:
- Paclitaxel and docetaxel are taxanes that impact microtubule polymerization.
- Observed differences in tumor sensitivity to taxanes suggest varying mechanisms of action.
- Docetaxel demonstrated superior efficacy over paclitaxel in inhibiting in vivo growth of human lung and prostate cancer models, but not breast cancer models.
Purpose of the Study:
- To compare the in vitro cytotoxicity of paclitaxel and docetaxel across various human cancer cell lines.
- To investigate the effects of these taxanes on beta-tubulin isoforms, apoptosis markers, and proteomic profiles.
- To identify potential biomarkers predicting differential sensitivity to taxane-based chemotherapy.
Main Methods:
- In vitro assessment of drug cytotoxicity in human prostate (LNCaP), lung (SK-MES, MV-522), and breast (MCF-7, MDA-231) cancer cell lines.
- Analysis of beta-tubulin isoform expression, apoptosis markers, and proteomic profiles.
- Correlation analysis between cytotoxicity, Bcl-2 expression, and in vivo drug sensitivity.
Main Results:
- Cytotoxicity varied across cell lines, with docetaxel generally being more effective.
- In vitro cytotoxicity directly correlated with Bcl-2 expression and inversely correlated with in vivo docetaxel sensitivity.
- Proteomic profiling revealed differential regulation of a specific protein by docetaxel and paclitaxel in lung cancer cells (SK-MES).
Conclusions:
- The enhanced activity of docetaxel in tumors with low Bcl-2 expression suggests its potential as a biomarker for drug sensitivity.
- Further research is warranted to explore Bcl-2 as a predictive biomarker for taxane therapy.
- Investigating docetaxel in combination with agents that reduce Bcl-2 expression may improve treatment outcomes for specific cancers.

Attenuation
When constant series resistance and shunt conductance are present, voltage and current equations are modified. The propagation constant indicates that voltage and current waves consist of both forward and backward traveling components. These waves attenuate as they propagate, with the attenuation factor related to the resistance and conductance. In a...
