Nucleic Acids

49.8K
Nucleic acids are the most important macromolecules for the continuity of life. They carry the cell's genetic blueprint and carry instructions for its functioning.
DNA and RNA
The two main types of nucleic acids are deoxyribonucleic acid (DNA) and ribonucleic acid (RNA). DNA is the genetic material in all living organisms, ranging from single-celled bacteria to multicellular mammals. It is in the nucleus of eukaryotes and in the organelles, chloroplasts, and mitochondria. In prokaryotes,...

Biosynthesis of Nucleic Acids

1.0K
Nucleic acid biosynthesis is a fundamental biochemical process that produces the purine and pyrimidine nucleotides essential for DNA and RNA synthesis. This pathway maintains a balanced nucleotide pool, preventing imbalances that could jeopardize genetic integrity and cellular function. Given the crucial role of nucleotides, their synthesis is tightly regulated to ensure proper cellular homeostasis.Purine BiosynthesisThe biosynthesis of purine nucleotides begins with ribose-5-phosphate, a...

Nucleic Acid Structure

8.4K
The pentose sugar in DNA is deoxyribose, while in RNA the pentose sugar is ribose. The difference between the sugars is the presence of the hydroxyl group on the ribose's second carbon and a hydrogen on the deoxyribose's second carbon. The phosphate residue attaches to the hydroxyl group of the 5′ carbon of one sugar and the hydroxyl group of the 3′ carbon of the sugar of the next nucleotide, which forms  a 5′ to 3′ phosphodiester linkage.

Area of Science:

  • Biotechnology
  • Molecular Diagnostics
  • Nanotechnology

Background:

  • Direct microRNA (miRNA) analysis in complex biological samples faces challenges like low sensitivity, specificity, and analyte absorption.
  • Current detection methods often require complex procedures such as enzymatic amplification, hindering rapid diagnostics.

Purpose of the Study:

  • To develop a novel assay for direct, sensitive, and specific quantification of miRNAs in complex media.
  • To create a simplified, single-step detection method for early cancer diagnosis.

Main Methods:

  • Fabrication of erythrocyte membrane-biointerfaced spherical nucleic acids (EMSNAs).
  • Utilizing EMSNAs for tailor-engineered signal amplification and anti-interference properties.
  • Single-step quantification of miRNAs without enzymatic amplification.

Main Results:

  • Achieved ultrasensitive quantification of miR-141 with a limit of detection down to 33.9 aM.
  • Demonstrated high selectivity in discriminating between miR-200 family members.
  • Successfully performed robust miRNA analysis in human serum, differentiating cancer patients from healthy donors.

Conclusions:

  • The developed EMSNA assay offers a simplified, rapid, sensitive, and specific method for miRNA detection.
  • The assay shows significant potential for the early diagnosis of cancers like lung and prostate cancer through direct analysis in human serum.
  • This approach may satisfy clinical diagnostic requirements for complex biological samples.
