Synovium-derived stromal cells (SDSCs) from osteoarthritis (OA) patients promote osteoclast formation more than those from healthy individuals. Telocytes (TCs) were also more prevalent in OA samples, suggesting their role in OA.
Area of Science:
- Cell Biology
- Immunology
- Rheumatology
Background:
- Mesenchymal stem/stromal cells (MSCs) are implicated in osteoarthritis (OA) pathophysiology.
- Telocytes (TCs) are stromal cells with unclear roles in OA.
Purpose of the Study:
- To investigate the role of synovium-derived stromal cells (SDSCs) in OA.
- To compare SDSCs from healthy and OA subjects.
- To explore the presence and potential role of TCs in OA.
Main Methods:
- In vitro comparison of SDSCs from healthy and OA subjects.
- Analysis of SDSC phenotype, morphology, and differentiation potential.
- Co-culture assays with Peripheral Blood Mononuclear Cells (PBMCs).
- Histological, immunohistochemical, ultrastructural, qRT-PCR, and functional assays.
Main Results:
- Both healthy and OA SDSCs stimulated osteoclast formation from PBMCs.
- OA-derived SDSCs were significantly more effective in promoting osteoclast-like cell formation.
- Telocytes (TCs) were more abundant in OA subject cultures, suggesting their involvement in OA.
Conclusions:
- SDSCs contribute to OA pathogenesis by promoting osteoclastogenesis.
- The increased presence of TCs in OA suggests a potential role in the disease.
- These findings support a multifaceted role for MSCs in OA, impacting treatment strategies.

Cell Potential and Free Energy
Thermodynamics is the branch of physics dealing with the relationship between heat and other forms of energy. In an electrochemical cell, chemical energy is converted into electrical energy.
Thus, a link can be predicted between cell potential, free energy change, and the equilibrium constant for the reaction. Cell potential can also be measured as the oxidant or the reducing strength, and similar acid-base strength measures are reflected in equilibrium...
Potential Energy
Chemical bonds that form attractive forces between atoms also contain potential energy, called chemical energy. When a chemical reaction...
