Receptor Tyrosine Kinase-Like Orphan Receptors 1 and 2 (ROR1 and ROR2) are promising targets for neuroblastoma (NBL) immunotherapy. Targeting ROR1 with antibodies enhanced natural killer cell cytotoxicity, suggesting a new treatment avenue for NBL relapse.
Area of Science:
- Immunology
- Oncology
- Molecular Biology
Background:
- Neuroblastoma (NBL) relapse is a major cause of mortality in high-risk patients despite advances in immunotherapy.
- Novel tumor-associated antigens are needed to improve immunotherapy efficacy and minimize toxicity.
- Receptor Tyrosine Kinase-Like Orphan Receptors 1 and 2 (ROR1 and ROR2) are expressed in various cancers with limited healthy tissue expression.
Purpose of the Study:
- To investigate ROR1 and ROR2 as potential immunotherapy targets in NBL.
- To evaluate the role of ROR1 and ROR2 in NBL migration and proliferation.
- To explore the efficacy of anti-ROR1 and anti-ROR2 immunotherapies for NBL.
Main Methods:
- Characterization of ROR1 and ROR2 mRNA and protein expression in NBL cell lines and patient samples.
- In vitro cytotoxicity assays using NK92 effector cells against NBL.
- Correlation analysis of ROR1/ROR2 expression with patient survival and prognosis.
Main Results:
- ROR1 and ROR2 are expressed across all stages of NBL.
- ROR1/ROR2 expression correlates with survival and prognosis in non-MYC amplified tumors.
- Anti-ROR1 antibody pretreatment enhanced NK92 cell-mediated cytotoxicity against NBL cells.
Conclusions:
- ROR1 and ROR2 represent novel targets for NBL immunotherapy.
- Combining anti-ROR1 antibodies with immune effectors like NK92 cells shows potential for off-the-shelf immunotherapy.
- Further research is warranted to explore combination therapies for ROR1-expressing malignancies.

Targets for Drug Action: Overview
Receptors are either membrane-spanning or intracellular proteins, which upon binding a ligand, get activated and transmit the signal downstream to elicit a response. Drugs bind receptors, either mimicking the action of endogenous ligands or blocking the receptor activity to bring about a modified response. Nearly 35% of approved drugs target the G...
Target Cell Response to Hormones
Notably, the cellular response can be regulated by altering the number of receptors expressed in the cell. For example, prolonged exposure to elevated hormone levels results in a gradual decline or down-regulation in the number of receptors for that specific hormone on the cell surface. Conversely, in response to low hormone levels, cells may use up-regulation, producing an...
