This study found lower levels of certain cytokines, like interleukin-6 (IL-6) and interleukin-10 (IL-10), in women with fibromyalgia (FM). These findings suggest a potential role for reduced immune signaling in FM pathophysiology.

- Serum samples were analyzed from 67 female FM patients and 35 healthy women.

- Fibromyalgia Severity Score, Widespread Pain Index (WPI), Symptom Severity Scale (SSS), Hospital Anxiety and Depression Scale (HADS-A), and Perceived Stress Scale (PSS-10) were assessed.

Main Results:
- Fibromyalgia patients exhibited higher clinical rating scales compared to controls.
- After adjusting for age, BMI, and comorbidities, FM patients had lower levels of IL-6, IL-10, and CXCL-8 compared to healthy controls.
- hs-CRP levels did not differ significantly between groups.
- Lowered IL-10, poorer sleep quality, reduced aerobic activity, higher anxiety, and more comorbidities were associated with FM diagnosis.
- Neural network analysis indicated sleep quality, PSS-10, HADS-A, and cytokines predicted WPI, while PSS-10, HADS-A, and IL-10 predicted SSS.
Conclusions:
- Reduced levels of IL-6 and IL-10 are independently associated with fibromyalgia in women, irrespective of confounders.
- These findings suggest that diminished IL-6 and IL-10 signaling may contribute to the pathophysiology of fibromyalgia.
- Immune dysregulation, particularly involving specific cytokine pathways, warrants further investigation in FM.
