Pharmaceutical Equivalents

183
As defined by regulatory standards, pharmaceutical equivalents require generic drug products to have identical dosage forms and chemically identical active pharmaceutical ingredients (APIs). They must adhere to compendial or applicable standards for potency, content uniformity, disintegration times, and dissolution rates. In the case of modified-release dosage forms, variations in drug content are permissible as long as the delivered amount remains consistent with the innovator drug product.

Area of Science:

  • Pharmacology
  • Materials Science
  • Drug Delivery Systems

Background:

  • Curcuminoids exhibit potent antioxidant, anti-inflammatory, and antibacterial properties.
  • Poor solubility, oxidative degradation, and low bioavailability limit curcuminoid therapeutic applications.
  • Overcoming these limitations is crucial for effective oral and topical drug delivery.

Purpose of the Study:

  • To develop an efficient microencapsulation system for curcuminoids.
  • To improve the solubility, stability, and bioavailability of curcuminoids.
  • To create a controlled-release topical delivery system for curcuminoids.

Main Methods:

  • Complex coacervation microencapsulation of curcuminoids using gelatin B and chitosan (30:1% w/w).
  • Characterization of curcuminoids microcapsules (CPM) for shape, flowability, and stability.
  • Evaluation of curcuminoid solubility and drug release kinetics post-microencapsulation and cross-linking.

Main Results:

  • Spherical, discrete, and free-flowing CPM with enhanced integrity and stability were produced.
  • Microencapsulation significantly increased curcuminoid solubility.
  • Chemical cross-linking facilitated sustained drug release, following zero-order kinetics with super case-II transport.

Conclusions:

  • Curcuminoids microcapsules (CPM) represent an effective strategy for enhancing curcuminoid delivery.
  • The developed CPM system improves solubility and provides controlled release, particularly for topical applications.
  • CPM incorporation optimizes the pharmacological benefits of curcuminoids for more efficacious drug delivery.
