Purpose of the Study:

  • To validate a rapid sampling microdialysis technique for measuring free cortisol.
  • To assess the dynamic changes in free and total cortisol in subcutaneous tissue and blood.
  • To compare microdialysis-derived free cortisol measurements with established methods.

Main Methods:

  • Healthy male volunteers received placebo, Synacthen, or dexamethasone.
  • Microdialysis was used for 10-minute sampling of free and total cortisol in subcutaneous tissue, blood, and saliva.
  • Measurements were taken for 1 hour before and 4 hours after administration.

  • Synacthen stimulation significantly increased total and free cortisol levels, peaking between 2-3 hours.
