Plasmacytoid dendritic cells (pDCs) play a role in bacterial infections. This study shows pDCs can evoke a Salmonella-specific cytotoxic T lymphocyte (CTL) response during S. Typhimurium infection.

Purpose of the Study:
- To analyze the Salmonella-specific cytotoxic T lymphocyte (CTL) response mediated by pDCs.
- To elucidate the role of pDCs in adaptive immunity against Salmonella Typhimurium.

- Plasmacytoid dendritic cells were stained with carboxyfluorescein succinimidyl ester (CFSE).
- Cells were pulsed with Salmonella Typhimurium antigens (OmpC73, Kb-restricted epitopes) or whole bacteria.
- Cytotoxicity assays were performed to measure CTL activity against target cells.
Main Results:
- A statistically significant specific lysis was observed when pDCs were pulsed with OmpC73 antigen.
- Significant CTL activity was also noted when pDCs were exposed to S. Typhimurium, particularly at a 1:100 effector:target ratio with OmpC73.
Conclusions:
- Plasmacytoid dendritic cells are capable of evoking a CTL response during Salmonella Typhimurium infection.
- pDCs contribute to the cellular immune response against Salmonella.
