Antibody Structure01:10

Antibody Structure

65.3K
Overview
Antibodies, also known as immunoglobulins (Ig), are essential players of the adaptive immune system. These antigen-binding proteins are produced by B cells and make up 20 percent of the total blood plasma by weight. In mammals, antibodies fall into five different classes, which each elicits a different biological response upon antigen binding.

Antibody Structure and Classes

8.4K
Antibodies, also known as immunoglobulins, are produced by B cells in response to foreign substances, such as bacteria and viruses. These proteins are critical for recognizing and neutralizing these substances, protecting the body from potential harm.
The basic structure of an antibody consists of four protein chains: two identical heavy chains and two identical light chains. These chains are held together by disulfide bonds and other non-covalent interactions, forming a Y-shaped structure.

Area of Science:

  • Biochemistry
  • Structural Biology
  • Immunology

Background:

  • Antibody and nanobody therapeutics are crucial in modern medicine.
  • Access to structural and sequence data for these biologics is vital for development and understanding.
  • Existing databases may not comprehensively link therapeutic approvals with detailed structural information.

Purpose of the Study:

  • To develop and present the Therapeutic Structural Antibody Database (Thera-SAbDab).
  • To provide a centralized resource linking World Health Organisation (WHO)-approved antibody therapeutics with structural data.
  • To facilitate sequence and structural analysis of therapeutic antibodies.

Main Methods:

  • Integration of WHO-recognized antibody and nanobody therapeutics.
  • Cross-referencing therapeutic data with the Structural Antibody Database (SAbDab) for variable domain sequence matches.
  • Weekly synchronization with SAbDab to incorporate new Protein Data Bank entries and sequence data.
  • Development of query functionalities based on therapeutic name, metadata, and sequence identity.

Main Results:

  • Thera-SAbDab provides comprehensive tracking of antibody and nanobody therapeutics.
  • Structural coverage, sequence alignments, and metadata are available for each therapeutic.
  • The database enables searching by therapeutic name, metadata, or variable domain sequence.
  • As of August 5, 2019, 461 unique therapeutic molecules were cataloged.

Conclusions:

  • Thera-SAbDab serves as a valuable, up-to-date resource for researchers studying therapeutic antibodies.
  • The database enhances the ability to analyze sequence-structure relationships in antibody therapeutics.
  • It supports drug discovery and development by providing accessible, integrated data.
