Summary
We identified 38 micro RNA (miRNA) expression quantitative loci (eQTL) in mouse brain tissue. This catalog provides insight into genetic regulation of miRNA and potential roles in brain function and disease.
Area of Science:
- Neuroscience
- Genetics
- Molecular Biology
Background:
- Micro RNAs (miRNAs) are key gene expression regulators influencing traits and diseases.
- Expression quantitative loci (eQTL) analysis links genetic variation to miRNA expression.
- Limited eQTL studies exist for miRNA in brain tissue.
Purpose of the Study:
- To catalog miRNA eQTL in mouse brain tissue.
- To characterize genetic influences on miRNA expression in naïve brain samples.
- To explore the biological mechanisms underlying miRNA regulation in the brain.
Main Methods:
- Utilized miRNA expression data from 881 miRNAs and 1416 genomic locations in a recombinant inbred mouse panel.
- Employed permutation p-values and zero permutation p-value correction for multiple testing.
- Conducted hotspot and Bayesian network analyses to investigate miRNA regulatory networks.
Main Results:
- Identified 38 significant miRNA eQTL after multiple testing correction.
- Ten miRNAs targeted genes enriched in brain-related pathways and mapped to four eQTL hotspots.
- Discovered four biological networks linking genetic variation, miRNA, and mRNA expression via Bayesian analysis.
Conclusions:
- Provided a comprehensive evaluation of miRNA eQTL in brain tissue.
- Identified candidate miRNAs for future research on brain regulation and disease.
- Highlighted the utility of miRNA eQTL for understanding genetic influences on brain function.

MicroRNA (miRNA) are short, regulatory RNA transcribed from introns (non-coding regions of a gene) or intergenic regions (stretches of DNA present between genes). Several processing steps are required to form biologically active, mature miRNA. The initial transcript, called primary miRNA (pri-mRNA), base-pairs with itself, forming a stem-loop structure. Within the nucleus, an endonuclease enzyme, called Drosha, shortens the stem-loop structure into hairpin-shaped pre-miRNA. After the pre-miRNA...

Regulation of Expression at Multiple Steps
1.3K
The gene expression in cells is regulated at different stages: (i) transcription, (ii) RNA processing, (iii) RNA localization, and (iv) translation. Transcriptional regulation is mediated by regulatory proteins such as transcription factors, activators, or repressors—these control gene expression by initiating or inhibiting the transcription of genes. Once a precursor or pre-mRNA is produced, it undergoes post-transcriptional modification, including 5' capping, splicing, and the...
