Area of Science:

  • Microbiology
  • Bacteriology
  • Genomics

Background:

  • The genus Acinetobacter includes important opportunistic pathogens.
  • Accurate identification and characterization of novel Acinetobacter species are crucial for understanding microbial diversity and potential health implications.

Purpose of the Study:

  • To isolate and characterize a novel bacterial strain from chicken meat.
  • To determine the taxonomic position of the novel strain using polyphasic characterization, including genomic and chemotaxonomic analyses.

Main Methods:

  • Isolation and cultivation of bacteria from chicken meat.
  • Gram staining, motility tests, and determination of optimal growth conditions (temperature, pH).
  • 16S rRNA gene sequencing, Average Nucleotide Identity (ANI), digital DNA-DNA hybridization (dDDH), and analysis of genomic DNA G+C content.
  • Chemotaxonomic analysis including respiratory quinone, cellular fatty acids, and polar lipid profiling.

Main Results:

  • A novel Gram-negative, milky-yellow, non-motile coccobacillus, designated S23T, was isolated.
  • Phylogenetic analysis based on 16S rRNA gene sequences placed strain S23T within the genus Acinetobacter, closely related to Acinetobacter defluvii.
  • ANI and dDDH values were below the thresholds for species delineation, supporting its novelty.
  • Genomic DNA G+C content was 41.53 mol%, with Q-9 as the major respiratory quinone, and specific profiles for major cellular fatty acids and polar lipids.

Conclusions:

  • Strain S23T represents a novel species within the genus Acinetobacter.
  • The proposed name for this new species is Acinetobacter pullicarnis sp. nov., with strain S23T as the type strain.
