Very small embryonic-like stem cells (VSELs) regenerate gonads and differentiate into gametes. These quiescent cells survive cancer therapy, offering hope for cancer survivors and potentially eliminating the need for gonadal tissue banking.
Area of Science:
- Reproductive Biology
- Stem Cell Science
- Oncology
Background:
- Very small embryonic-like stem cells (VSELs) are a novel stem cell population found in adult testes and ovaries.
- VSELs are developmentally linked to spermatogonial stem cells (SSCs) and ovarian stem cells (OSCs), originating from primordial germ cells (PGCs).
- VSELs are quiescent and small, undergoing asymmetrical cell divisions to produce SSCs/OSCs.
Purpose of the Study:
- To elucidate the role and potential applications of VSELs in reproductive medicine and oncology.
- To investigate the differentiation capacity of VSELs and their response to hormonal stimuli.
- To explore the possibility of using VSELs for gonad regeneration in patients with gonadal insufficiency, including cancer survivors.
Main Methods:
- In vitro culture of VSELs on feeder support without growth factors.
- Analysis of VSEL differentiation into sperm/oocyte-like structures.
- Assessment of VSEL expression of hormone receptors (FSHR, ER).
Main Results:
- VSELs differentiate into haploid sperm/oocyte-like structures in vitro.
- VSELs express FSH and estrogen receptors, indicating hormonal responsiveness.
- VSELs' quiescent nature allows survival through oncotherapy, suggesting potential for gonad regeneration.
Conclusions:
- VSELs can be utilized to regenerate gonads in cases of gonadal insufficiency, particularly in cancer survivors.
- VSELs are promising candidates for in vitro gamete differentiation.
- Understanding VSEL self-renewal is crucial for preventing testicular and ovarian cancers.
