Summary

This study investigated the antigenicity of muroctasin (MDP-Lys(L18)) in animal models. Muroctasin alone did not elicit significant antibody responses, but when conjugated with ovalbumin, it showed potential antigenic properties in guinea pigs and rabbits.

Background:

  • N2-[(N-acetylmuramoyl)-L-alanyl-D-isoglutaminyl]-N6-stearoyl-L-lysi++ + ne (MDP-Lys(L18)), also known as muroctasin, is a synthetic immunomodulator.
  • Understanding its antigenic potential is crucial for its development and application in immunology.

Purpose of the Study:

  • To evaluate the antigenicity of muroctasin (MDP-Lys(L18)) in mice, guinea pigs, and rabbits.
  • To determine if muroctasin alone or conjugated to ovalbumin (OA) can elicit immune responses.

Main Methods:

  • Passive cutaneous anaphylaxis (PCA), systemic anaphylaxis (SA), Arthus, and delayed skin reactions were employed.
  • Animals were sensitized with varying doses of MDP-Lys(L18) or MDP-Lys(L18)-OA conjugate using different adjuvants and routes.
