Neuritin (Nrn1) is expressed in various human tissues and is linked to 13 cancers. Downregulation of Nrn1 correlates with improved survival in most cancer patients, suggesting its potential as a biomarker.
Area of Science:
- Molecular Biology
- Oncology
- Biochemistry
Background:
- Neuritin (Nrn1) is a known central nervous system protein.
- Its expression outside the nervous system and role in human cancers remain largely uncharacterized.
Purpose of the Study:
- To investigate the expression of Nrn1 in diverse human tissues.
- To determine the association of Nrn1 with various human cancers.
Main Methods:
- Nrn1 gene expression analyzed using Clontech Multiple Tissue cDNA panel.
- Protein detection via immunohistochemistry; subcellular localization by immunocytochemistry and western blotting.
- Cancer expression data obtained from the online tool GEPIA.
Main Results:
- Nrn1 mRNA detected in placenta, lungs, skeletal muscle, thymus, pancreas, liver, and heart.
- Nrn1 localized to plasma membrane lipid rafts in SH-SY5Y cells.
- Nrn1 downregulated in 12 cancers (e.g., BLCA, BRCA, COAD) and upregulated in DLBC.
- Downregulation of Nrn1 correlated with longer survival in 12 tumor types, excluding DLBC.
Conclusions:
- Nrn1 is expressed in multiple human tissues, localized to cell membrane lipid rafts.
- Nrn1 is significantly associated with 13 human cancers.
- Nrn1 shows potential as a biomarker and therapeutic target for these cancers.

Enzyme-linked Receptors
Neurotrophin (NT) receptors are a family of RTKs, including trkA, trkB, and trkC (tropomyosin-related kinase) receptors. TrkA is specific for nerve growth factor (NGF), neurotrophin-6, and neurotrophin-7. TrkB binds...

The Retinoblastoma Gene
The first-ever tumor suppressor gene called Rb was identified in retinoblastoma - a rare eye tumor in children. In inherited forms of the disease, a child inherits one defective copy of the Rb gene, which predisposes them to retinoblastoma. However,...
