Cell separation was first achieved in 1964 by S. H. Seal, who separated large tumor cells from the smaller blood cells using filtration. Two years later, Pohl and Hawk performed experiments on how cells respond differently to a nonuniform electric field based on the cell type. Such observations were the inception of cell separation methods, which allow isolating a single cell type from a heterogeneous sample.

Background:

  • Cytology specimens are typically processed using cytocentrifugation, a method that, while preferred over membrane filters, is expensive and can cause cellular distortion.
  • Existing methods for processing clear cytology specimens are costly and may compromise cellular integrity.

Purpose of the Study:

  • To standardize and evaluate "Agar cell-suspension" (ACS), an innovative, simple, and cost-effective technique for processing clear cytology specimens.
  • To compare the efficacy of ACS with conventional cytocentrifugation in terms of cellularity, cytomorphology, staining, and cost.

  • A total of 93 clear specimens (urine, effusion, CSF, bronchial lavage) were processed using both cytocentrifugation and the novel ACS technique.
