Mismatch Repair

6.2K
Organisms are capable of detecting and fixing nucleotide mismatches that occur during DNA replication. This sophisticated process requires identifying the new strand and replacing the erroneous bases with correct nucleotides. Mismatch repair is coordinated by many proteins in both prokaryotes and eukaryotes.
The Mutator Protein Family Plays a Key Role in DNA Mismatch Repair
The human genome has more than 3 billion base pairs of DNA per cell. Prior to cell division, that vast amount of genetic...

Area of Science:

  • Oncology
  • Molecular Diagnostics
  • Genetics

Background:

  • Microsatellite instability (MSI) is a key biomarker in colorectal cancer (CRC).
  • Accurate MSI classification is crucial for understanding CRC phenotypes and guiding treatment.
  • Existing MSI analysis methods may have limitations in challenging cases.

Purpose of the Study:

  • To evaluate the effectiveness of the ProDx® MSI analysis system for classifying MSI in CRC.
  • To compare the ProDx® MSI system with established methods like Promega MSI analysis System 1.2 and the NCI panel.

Main Methods:

  • Compared MSI profiles of 97 CRC FFPE samples using ProDx® MSI, Promega MSI analysis System 1.2, and NCI panel.
  • Validated results with immunohistochemistry (IHC) for mismatch repair (MMR) protein expression.
  • Utilized next-generation sequencing for discordant results.

Main Results:

  • ProDx® MSI identified 3 additional MSI-High CRC cases missed by other systems.
  • ProDx® MSI demonstrated higher concordance with IHC (99.0%) compared to Promega (96.9%) and NCI (95.9%).
  • Achieved 100% sensitivity and 98.4% specificity for MSI status determination.

Conclusions:

  • ProDx® MSI system accurately classifies colorectal cancer MSI status.
  • This system reveals more MSI-High cases in CRC than previous methods.
  • The ProDx® MSI system offers improved diagnostic potential for MSI analysis in CRC.
